摘要: Publisher Summary This chapter presents a study on the three-dimensional dynamic structure of transfer RNA (tRNA) by nuclear magnetic resonance (NMR) spectroscopy. 13 C-enriched, native modified nucleosides are used as probes, whose aromatic and proton chemical shifts Nuclear Overhauser Effect (NOE) values give access to internucleotide distances bases not involved in Watson–Crick pairs. In Escherichia coli ( E. ) , tRNA multiple peaks for methyl groups T Phe Tyr Gm latter, indicates structural forms regions molecule probed. These coalesced single upon addition MgCl 2 . The studies carbon NMR signals yeast correlated with their corresponding resonances via two-dimensional carbon-proton correlation experiment. Although analysis protein perturbation conformation is difficult, other effectors site-specific quite successful.
