摘要: Purified preparations of circulating leukaemic blast cells from patients with acute myeloid (M1–7) or lymphoblastic leukaemia, and the lymphoid chronic lymphocytic forms were incorporated into clots prepared fibrinogen plasminogen. Patterns lysis followed measured by light transmission in a microtitre plate reader. Mature polymorphonuclear mononuclear cell fractions normal individuals studied concurrently for comparison. Blast leukaemia (M2–4) ‘myeloid’ capable lysing plasminogen-containing clots; this activity was neutralized addition immunoglobulin against urokinase plasminogen activator (u-PA), but not anti-tissue plasmogen (t-PA). lacked lytic activity, as did leukaemia. Lysed showed presence free on sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS–PAGE) overlay zymography, also anti-u-PA, whereas not. These observations suggest that mechanisms underlying some severe bleeding leukaemias have critical fibrinolytic component generated themselves.
