摘要: This study aimed at getting a deeper insight in the molecular mechanism by which natural furanone (5Z)-4-bromo-5-(bromomethylene)-3-butyl-2(5H)-furanone disrupts quorum sensing Vibrio harveyi. Bioluminescence experiments with signal molecule receptor double mutants revealed that blocks all three channels of V. harveyi system. In further using mutations transduction pathway, compound was found to block sensing-regulated bioluminescence interacting component located downstream Hfq protein. Furthermore, reverse transcriptase real-time polymerase chain reaction specific primers showed there no effect on luxR(Vh) mRNA levels wild-type cells. contrast, mobility shift assays presence furanone, significantly lower LuxR(Vh) response regulator protein were able bind its target promoter sequences Finally, tests purified also less shifts furanone-treated LuxR(Vh), whereas concentration not be altered (as determined SDS-PAGE). Therefore, our data indicate rendering master unable genes.
