Chemotherapy of Onchocerca lienalis microfilariae in mice: a model for the evaluation of novel compounds for the treatment of onchocerciasis.

摘要: The model of Onchocerca lienalis microfilariae (mf) injected into inbred CBA/Ca mice was studied for its usefulness as an additional primary/secondary drug screen onchocerciasis. Invermectin, DEC, suramin, flubendazole, mebendazole, levamisole, Mel W, furapyrimidone, metrifonate, amoscanate and the new Ciba-Geigy compounds CGP 6140, 20'376 CGI 17658 all significantly reduced levels mf at a dose 5 X 100 mg/kg or less. An early dosing protocol, on days 3-7 after infection, found to be generally more effective than 11-15, followed by necropsy day 18. In some cases there were important differences in activity depending whether administered subcutaneous oral route, indicating that should tested via both routes. Ivermectin far most active compound examined, virtually clearing from skin 0.0063 producing significant reduction (63.5%) 0.0008 following administration. comparison, DEC much less active, 32.4% 25 ranging up maximum 72% mg/kg. examined next ivermectin, almost 100% against 6.25 route while being administration (65% reduction). lowest 3.13 (per os) which 64%. also very resulting 46% (subcutaneous) 62% This mouse has clearly identified known microfilaricides also, lesser extent, those considered principally macrofilaricides. We believe it value onchocerciasis, will enable evaluation novel skin-dwelling level, providing complementary information vitro screens using adult Onchocerca.
