摘要: Histone acetylation has recently been implicated in learning and memory processes, yet necessity of histone for such processes not demonstrated using pharmacological inhibitors acetyltransferases (HATs). As such, the present study tested whether garcinol, a potent HAT inhibitor vitro , could impair hippocampal consolidation block memory-enhancing effects modulatory hormone 17β-estradiol (E 2 ). We first showed that bilateral infusion garcinol (0.1, 1, or 10 μg/side) into dorsal hippocampus (DH) immediately after training impaired object recognition ovariectomized female mice. A behaviorally effective dose (10 also significantly decreased DH activity. next examined subeffective (1 ng/side) E on epigenetic processes. Immediately training, mice received infusions vehicle, (5 μg/side), ng/side), plus garcinol. Forty-eight hours later, blocked . Garcinol reversed -induced increase H3 acetylation, activity, levels de novo methyltransferase DNMT3B, as well decrease repressor protein deacetylase 2. Collectively, these findings suggest is critical beneficial recognition. Importantly, this work demonstrates role can be studied inhibitor.
