Modulation of the Selectivity of Immobilized Lipases by Chemical and Physical Modifications: Release of Omega-3 Fatty Acids from Fish Oil

摘要: Three different lipases (from Candida antarctica fraction B (CALB), Thermomyces lanuginose (TLL), and Rhizomucor miehei (RML)) were immobilized by two methods, immobilization on CNBr-activated Sepharose via a mild covalent or adsorption onto hydrophobic supports (Octyl-Sepharose). These preparations chemically physically modified the protein surface (enzyme carboxylic groups with ethylenediamine, amino succinic anhydride, coating polyethyleneimine).The activity selectivity in production of eicosapentaenoic acid (EPA) docosahexaenoic (DHA) enzymatic hydrolysis sardine oil evaluated. Activity dependent lipases, protocols, modification pH reaction media. The (EPA/DHA ratio) RML was increased after succinylation from 7.5 to 34 at 6.0. octyl-RML improved 1.5 8.5 when 6 8. octyl-TLL twofold PEI 6. properties CAL-B derivatives slightly altered modification.
