Metallothionein-I/II Knockout Mice Aggravate Mitochondrial Superoxide Production and Peroxiredoxin 3 Expression in Thyroid after Excessive Iodide Exposure

摘要: Purpose. We aim to figure out the effect of metallothioneins on iodide excess induced oxidative stress in thyroid. Methods. Eight-week-old MT-I/II knockout (MT-I/II KO) mice and background-matched wild-type (WT) were used. Mitochondrial superoxide production peroxiredoxin (Prx) 3 expression measured. Results. In vitro study, more significant increases mitochondrial Prx detected KO groups. vivo significantly higher concentrations urinary iodine level 100 HI group. Compared NI group, there was no difference existing serum thyroid hormones either groups (P > 0.05), while increased with LDH activity decreased relative cell viability. WT mice, changes No differences between group 10 both 0.05). Conclusions. Iodide a without MT I/II protection may result strong stress, which further leads damage thyrocytes.
