Microelectrode studies on the acid microenvironment beneath adherent macrophages and osteoclasts.

摘要: Osteoclasts and activated macrophages in culture were shown to generate an acidic microenvironment specifically the attachment zone between cell base of dish. Measurements using pH microelectrodes revealed that osteoclasts, when firmly attached, could achieve a fall about 1 unit min-1 limit value 3.0 or less. Activated produced slower 0.5-2 units h-1 3.6-3.7 was generally detected. The method activation relatively unimportant, but where formed clumps effect reinforced. also examined situ osteoporotic bone fragments rabbit ear chambers, combination H+ Ca2+ double-barrelled electrode. readings reached lower 4.7 calcium concentration rose maximum 40 mM erosion sites. In vivo such acid conditions would favor direct extracellular action secreted lysosomal proteinases degradation collagen by both types.
