High expression of cloned immunoglobulin k gene in transgenic mice is restricted to B lymphocytes

摘要: Immunoglobulin genes are normally expressed only in cells of the B lymphocyte lineage after a variable (V) and constant (C) gene rearrangement has occurred1. To study control immunoglobulin expression defined situation, we have produced transgenic mice by microinjecting rearranged mouse k (designated pB1-14) into fertilized eggs2. We present here analysis six different k-transgenic lines. All express microinjected completely tissue-specific fashion. Transcripts from pB1-14 found at high level spleen, but undetectable nonlymphoid tissues testis, liver, kidney, heart, muscle, brain thyroid gland. In lymphoid cell subpopulations, transcripts is correlated with relative number cells; there no correlation proportion T lymphocytes. concluded, therefore, that contains target sequences for lymphocyte-specific activation signals override influence integration site.
