摘要: Circulating cell-free(cf) microRNAs (miRNAs) have been reported to exist in plasma. MicroRNA-210(miR-210) is known play important roles the tumor hypoxic state. We hypothesized that expression levels of cf-miR-210 plasma would predict early clinical recurrence melanoma patients. A direct miRNA assay on (RT-qPCR-DP) was developed improve cf-miRNA logistics, eliminate RNA extraction, and reduce specimen amount required. extracted from formalin-fixed paraffin-embedded (FFPE) tissues (n = 108) assessed by RT-qPCR. Plasma (10 μl; n 264) procured AJCC Stage III/IV patients phase III trials. RT-qPCR-DP performed detect cf-miR-210. MiR-210 significantly higher metastatic tumors compared primary tumors. Cf-miR-210 versus healthy donor controls. In serial bloods within individual patients, < 3 months prior disease increased baseline (p 0.012). ROC curve analysis demonstrated with elevated were more likely recurrence. Moreover, increase correlated poorer prognosis 0.001). Lactate dehydrogenase (LDH) level also AIC values for proportional hazards regression models cf-miR-210(120.01) LDH (122.91) a better indicator. concluded enhanced provides identification systemic
