摘要: Parkinson's disease (PD) is characterised by the preferential loss of dopaminergic neurones from substantia nigra (SN) that leads to hallmark motor disturbances. Animal and human studies suggest a beneficial effect oestrogen nigrostriatal system, regulation neurotrophic factor expression oestrogens has been suggested as possible mechanism contributing neuroprotective effect. The present study was designed investigate whether neuroprotection exerted 17β-oestradiol on mediated through glial cell line-derived (GDNF) expression. Using an in vivo rat model PD, we were able confirm relevance defending against 6-hydroxydopamine (6-OHDA) toxicity. 17β-oestradiol, released micro-osmotic pumps, implanted 10 days before intrastriatal 6-OHDA injection, prevented induced 6-OHDA. treatment also promoted increase GDNF protein levels both SN striatum. To explore increases neuroprotection, analysed, neurone-glia cultures, antibody neutralisation RNA interference-mediated knockdown. results showed silencing abolished protection provided Taken together, these strongly identify important player 17β-oestradiol-mediated neuroprotection.
