摘要: MRI enhanced with a macromolecular contrast medium (MMCM) has previously been shown to estimate tumor microvascular characteristics that correlate closely histologic density, an established surrogate of angiogenesis. A similar MMCM-enhanced technique now used investigate the acute effects antibody-mediated inhibition vascular endothelial growth factor (VEGF), well-studied and potent angiogenesis stimulator. Athymic rats xenografted human breast carcinoma (MDA-MB-435) were imaged after administration albumin-gadolinium diethylenetriamine pentaacetic acid (Gd-DTPA30) using heavily T1-weighted three dimensional-spoiled gradient-refocused acquisition in steady-state pulse sequence before 24 hours treatment anti-VEGF antibody (single dose 1 mg). Changes longitudinal relaxivity (delta R1) analyzed bidirectional two-compartment kinetic model fractional blood volume (fBV) permeability surface area product (PS). Data showed significant decrease (P < 0.05) PS respect at antibody. No change was observed fBV. Suppression induced by can be detected quantified MRI. grading monitoring anti-angiogenesis interventions could find wide clinical application.
