摘要: In common with many addictive substances and behaviors nicotine activates the mesolimbic dopaminergic system. Brain microdialysis studies in rodents have consistently shown increases extrasynaptic DA levels striatum after administration of but PET experiments primates given contradicting results. A recent study assessing effect smoking humans showed no change ( 11 C)raclopride binding brain, did find that ''hedonia'' correlated a reduction suggesting may mediate posi- tive reinforcing effects nicotine. this experiment we measured nico- tine, administered via nasal spray, on release using PET, 10 regular smokers. There was overall any striatal regions examined. However, individual subjective measures ''amused'' ''happiness'' associative (AST) sensorimotor stria- tum (SMST). Nicotine concentration negatively BP limbic striatum. had significant cardiovascular including pulse rate, systolic blood pressure (BPr), diastolic BPr. Baseline potential (BP) AST Fagerstrom score, an index dependence. These results support role for system tine addiction, reveal more complex relationship than suggested by animals.
