摘要: alpha,beta-Unsaturated aldehydes were investigated in vitro for their ability to inhibit superoxide anion radical (O2-.) production stimulated human polymorphonuclear leukocytes (PMN). The (i) trans-4-hydroxynonenal and malonaldehyde (MDA), two toxic lipid peroxidation products; (ii) acrolein crotonaldehyde, air pollutants derived from fossil fuel combustion; (iii) trans,trans-muconaldehyde, a putative hematotoxic benzene metabolite. Preincubation of PMN with reactive followed by stimulation the oxygen burst initiator phorbol myristate acetate (PMA) resulted dose-dependent inhibition O2-. production. concentration at which 50% (IC50) was observed 21 microM acrolein, 23 27 330 crotonaldehyde. A similar inhibitory effect these digitonin- concanavalin A-stimulated PMN. MDA inhibited PMA-stimulated 100% 10(-2) M but gave no 10(-3) M. standard aldehyde propionaldehyde did not 10(-3)-10(-6) presence cysteine completely protected against this aldehyde. results indicate that correlates directly alkylation potential is function electrophilicity beta carbon.
