摘要: Role of renal endopeptidase 24.11 in kinin metabolism vitro and vivo. The relative contributions three kininases to total urinary kininase activity were determined by measuring the hydrolysis kinins presence absence inhibitors I (2-mercapto-methyl-3-guanidinoethylthiopropanoic acid; MGTA), II (cap-topril) neutral (NEP or enkephalinase A; phosphoramidon). Surprisingly, NEP was responsible for 68 ± 2% (N = 18) rat while contributed only 9 0.4% 23 1%, respectively. To study effects inhibition on function, phosphoramidon (110 330 µg/hr/kg; N=6) saline (0.1 µN/min; infused into rats. Urinary kinins, kininases, blood flow (RBF), glomerular filtration rate (GFR), U Na V, K V UV measured during control, experimental recovery periods. Phosphoramidon at higher dose decreased from 284 49 58 5 ng/min/kg (77%, P 12 2 17 3 µEq/min/kg (37%, did not change. 125 I-Tyr 0 -bradykinin aorta appear urine intact simultaneous captopril adminstration. This is first demonstration having a major role catabolism kinins. increase after administration may be due intrarenal destruction.
