摘要: Common hosts for the large-scale manufacture of biological products, such as Escherichia coli and Saccharomyces cerevisiae, do not excrete products to medium. Effective techniques cell disruption are therefore required. These include physical, chemical, enzymatic mechanical methods. Mechanical methods bead milling, high-pressure homogenization, microfluidization preferred. However, gentler, specific receiving increasing attention particularly when used in combination synergistically exploit their different specificities. Benefits can also be derived by integrating product release recovery. In all cases it is essential consider interaction operation with downstream units clearly demonstrate cost benefits alternative strategies.
