摘要: p27Kip1 (p27) is a critical cyclin dependent kinase inhibitor that negatively regulates cell cycle progression from G1 to S phase. Levels of nuclear p27 are reduced in many human cancers. Both the levels and function regulated by transcriptional, translational, posttranslational mechanisms. In regulating p27, phosphorylation protein particularly important for determining susceptibility ubiquitination proteolytic degradation, as well levels, subcellular localization, functional activity p27. addition control cycle, cytoplasmic expression can enhance motility, increase renewal stem cells, inhibit apoptosis, induce autophagy, stimulate tumor metastasis. Persistence provides an independent favorable prognostic factor Expression or induction may provide predictive biomarker response chemotherapy, hormonal therapy, at least some targeted therapies including trastuzumab Src inhibitors. addition, restoration might be achieved pharmacologically, inhibiting proliferation cancer cells. Inhibition SKP2-mediated one additional promising approach.
