Stability of potato (Solanum tuberosum L.) plants regenerated via somatic embryos, axillary bud proliferated shoots, microtubers and true potato seeds: a comparative phenotypic, cytogenetic and molecular assessment.

摘要: The stability, both genetic and phenotypic, of potato (Solanum tuberosum L.) cultivar Desiree plants derived from alternative propagation methodologies has been compared. Plants obtained through three clonal routes-axillary-bud-proliferation, microtuberisation a novel somatic embryogenesis system, true seeds (TPS) produced by selfing were evaluated at levels: gross phenotype minituber yield, changes in ploidy (measured flow cytometry) molecular marker analysis [measured using AFLP (amplified fragment length polymorphism)]. clonally propagated exhibited no phenotypic variation while the TPS-derived showed obvious segregation. Significant differences observed with respect to yield average plant height, time harvesting, was not significantly different among four routes. None types varied genome constitution as assessed cytometry. However, very low level profile seen amongst embryo (3 out 451 bands) microtuber (2 plants. Intriguingly, only markers generated methylation sensitive restriction enzymes found show polymorphism. No polymorphism regenerated axillary-bud-proliferation. could be significant on genome-wide scale, is discussed context possible occurring during process embryogenesis.
