摘要: Abstract Rationale. Cocaine is often abused in a "binge" pattern, but little known about changes plasma cocaine concentrations or pharmacokinetics during administration of multiple doses. Moreover, the extent to which gender may influence levels binge has not been studied rhesus monkeys. Objectives. To compare effects repeated injections same dose (0.4 mg/kg 0.8 mg/kg, i.v.) on concentrations, and behavioral responses male female Methods. Four were administered at 30-min intervals five six Samples for analysis collected 2, 4, 8, 16 24 min after first three injections. After fourth injection, additional samples 10-min over 150 min. Results. Plasma peaks nadirs increased monotonically successive (P<0.0001). Peak measured 2 min higher 0.8 mg/kg than 0.4 mg/kg (P<0.006). There no significant differences time peak (tmax), (Cmax) half-life (t1/2; min) either cocaine. Group average ratings similar males females each Conclusions. progressively low high episodes, there pharmacokinetics. These findings demonstrate feasibility simulating patterns
