Quantitative-profiling of bile acids and their conjugates in mouse liver, bile, plasma, and urine using LC-MS/MS

摘要: The differences among individual bile acids (BAs) in eliciting different physiological and pathological responses are largely unknown because of the lack valid simple analytical methods for quantification BAs their taurine glycine conjugates. Therefore, a sensitive LC-MS/MS method simultaneous 6 major BAs, glycine, conjugates mouse liver, bile, plasma, urine was developed validated. One-step sample preparation using solid-phase extraction (for urine) or protein precipitation plasma liver) used to extract BAs. This is with limit ranging from 10 40 ng/ml various analytes, has large dynamic range (2500), short run time (20 min). Detailed BA profiles were obtained this method. Muricholic acid (MCA) cholic (CA) constituted more than 90% liver bile. concentrations about 300-fold higher 180-fold liver. In summary, reliable quantify metabolites applied tissues fluids.
