Autocrine Growth Regulation in Fetal and Adult Human Fibroblasts

摘要: Abstract Medium conditioned (CM) by human fetal fibroblasts stimulates proliferation in sparse cultures. This effect is inhibited suramin and staurosporine, indicating the presence of autocrine growth factors CM. On contrary, CM inhibits DNA synthesis confluent cultures, suggesting a regulatory role for secreted factors. The profile persists during vitro ageing. However, it changes dramatically fetal-to-adult transition, as adult are stimulated CM, regardless culture density. These effects similar to those that TGF-β known have on fibroblasts. Indeed present media fibroblasts, but CM-activity cannot be ascribed solely this factor. Fibroblasts originating from different tissues exhibit same features, general character mechanism.
