Citrobacter rodentium Infection in Mice Elicits a Mucosal Th1 Cytokine Response and Lesions Similar to Those in Murine Inflammatory Bowel Disease

摘要: Citrobacter rodentium is a classically noninvasive pathogen of mice that similar to enteropathogenic Escherichia coli (EPEC) in man. Following oral infection young mice, the organism colonizes distal colon, and within 1 week colonic mucosa doubles thickness there massive epithelial cell hyperplasia. Since T-cell responses mouse models inflammatory bowel disease (IBD) also cause hyperplasia, we have investigated possibility C. promotes mucosa, thereby increasing shedding, transmission, replication organism. Beginning 6 days after infection, bacteria were observed be close association with surface visible scattered throughout lamina propria submucosa. There was CD3+-cell infiltrate into epithelium as well mucosal thickening crypt The majority CD3+ cells CD4+ not γδ+. Reverse transcription-PCR analysis cytokines revealed highly polarized Th1 response (interleukin-12, gamma interferon, tumor necrosis factor alpha) large increase mitogen keratinocyte growth factor. None changes seen inoculated lacking intimin (which necessary for colonization), but they complemented from EPEC. This first example bacterial which elicits strong produces pathology IBD, characterized by response. These results suggest responds stereotypic way responses.
