摘要: Simvastatin has been shown to stimulate osteogenesis both in vitro and vivo. However, the mechanism by which simvastatin exerts its effects is still unclear. We previously reported that promotes bone morphogenetic protein 2 (BMP-2) expression, induces osteoblastic differentiation, inhibits adipocytic differentiation mouse marrow stromal cells (BMSCs), this occurs, at least part, via a BMP-2-dependent pathway. The aim of study was investigate further mechanisms stimulates BMSCs. To determine whether simvastatin-mediated dependent on BMP-2, BMSCs were treated with nonimmune normal IgG or BMP-2 neutralizing antibodies combined different concentrations simvastatin. Surprisingly, stimulatory effect alkaline phosphatase (ALP) activity not completely blocked monoclonal antibody treatment. Interestingly, we found estrogen receptor-alpha (ER-α) levels increased after for 72 h concentration-dependent manner. Moreover, ALP receptor agonist ICI 182,780, cotreatment 17-β-estradiol activities two-to threefold compared treatment alone. These results suggest simvastatin-induced mediated, induction ER-α provide new insight into formation
