摘要: Subtilisin-like serine proteases make up one of the most important allergen-families regarding number individual allergens. Previously, fungal subtilisin-like have been identified from Aspergillus-, Penicillium-, and Trichophyton-species having a prevalence IgE-reactivity between 33% 80%. Since IgE-cross-reactivity is common phenomenon within species we wanted to know whether this protein also represents an allergen in Cladosporium herbarum. Hence, screening C. herbarum cDNA library was performed using coding sequence Penicillium oxalicum vacuolar protease (Pen o 18) as hybridization probe, ending with full-length clone. Biochemical immunological characterization clone revealed that likely synthesized precursor N-terminal pro-enzyme minor (Cla h 9) 15.5%. Furthermore Cla 9 specifically reacted two monoclonal antibodies FUM20 PCM39, do Aspergillus fumigatus species. Investigation other cross-reactivity higher than alkaline proteases. IgE-epitope mapping done order test four 9-peptides high homology previously determined Pen ch 18-IgE-epitopes harbour IgE-epitopes. Three-dimensional models chrysogenum were generated for three-dimensional localization 9- 18- IgE-reactive -non-reactive peptides. Taken together new has identified, which may be useful molecule-based approach allergy-diagnosis -therapy. Moreover, further member allergen-family, stresses importance these proteins respect IgE-cross-reactivity.
