Comparative transcriptome analysis of human conjunctiva between normal and conjunctivochalasis persons by RNA sequencing

摘要: Abstract Conjunctivochalasis (CCH) is a common ocular disease, especially in aged people. However, the molecular mechanism of CCH on transcriptional level has been unclear. In this study, we characterized landscape human conjunctiva and compared transcriptome between normal persons (n = 10) patients (n = 11). Illumina RNA sequencing (RNA-seq) was performed to obtain data, these data were analyzed using various bioinformatics methods, including read mapping, analysis gene expression, ontology (GO) Kyoto Encyclopedia Gene Genomes (KEGG) metabolic pathway analysis. Additionally, expression patterns 20 dysregupated genes validated by qRT-PCR. RNA-seq result showed that clean ratios 21 samples more than 95% 92% all reads (32–41 million reads) mapped genome sequence. There 175 up-regulated 582 down-regulated identified persons. Among CCH, many related with cell cycle proliferation, such as BUB1, CCNB1, CCNB2 CENPA, which might disturb growth proliferation. addition, several associated keratinization differentiation epidermal cells, SPRR1A, SPRR1B, CALML5. over-expressed genes, CALML6 play important roles development CCH. The results qRT-PCR confirmed accuracy credibility RNA-Seq This study provided lot valuable information about pathogenic could be used better future.
