摘要: Lifestyle, particularly smoking and alcohol consumption, may induce and/or inhibit drug metabolism. In order to reveal the effects of consumption on 5-fluorouracil (5-FU)-related metabolic enzymes, namely thymidylate synthase, dihydropyrimidine dehydrogenase (DPD; a sole catabolic enzyme 5-FU), orotate phosphoribosyl transferase (OPRT) thymidine phosphorylase, in oral squamous cell carcinomas, mRNA expression these enzymes was investigated 29 surgical specimens compared by Brinkman index drinking years. The were divided into normal tumor regions independently analyzed using quantitative reverse transcription-polymerase chain reaction. There significantly positive correlation between DPD tissues index/drinking years, with OPRT being correlated tissues. These results revealed that lifestyle habits, including vary activity 5-FU-related enzymes. is initial rate-limiting pathway 5-FU. Therefore, reduce anticancer 5-FU, possibly through induction activity.
