摘要: Abstract Elevated cell Na + -H exchange (NHE) activity characterizes diabetic nephropathy (DN), but the mechanisms of this abnormality are unclear. Recent evidence suggests that NHE and Ca 2+ pump share similar regulatory pathways, whether abnormalities in metabolism characterize DN is not known. We investigated efflux rates, activity, cytosolic ([Ca ] i ) concentrations, intracellular pH (pH human skin fibroblasts from 20 patients with type 1 (insulin-dependent) diabetes nephropathy; normoalbuminuria matched for age, sex, duration diabetes; 10 individuals without diabetes. pump-mediated was significantly lower than (0.074 ± 0.01 versus 0.115 0.131 0.02 nmol·mg protein −1 ·min ; analysis variance [ANOVA], P = 0.015). maximal velocity exchanger confirmed (14.4 1.2 7.1 0.7 8.0 mmol H ·l ANOVA, rates could be shown. Adjustment glycated hemoglobin plasma lipid levels did affect these findings. Finally, [Ca concentrations were normal all patients.
