Glycolytic enzyme expression and pyruvate kinase activity in cultured fibroblasts from type 1 diabetic patients with and without nephropathy

摘要: Abstract Since type 1 diabetes mellitus (T1DM) patients with nephropathy (DN+) are insulin-resistant, we aimed to identify (new) potential molecular sites involved in the alterations of glucose metabolism these patients. We examined expression glycolytic enzymes cultured fibroblasts from T1DM(DN+) as compared those T1DM without (DN−) and controls. Pyruvate kinase (PK) activity was also determined. Human skin were grown normal (6 mM). RNAs proteins analyzed, respectively, using cRNA microarray two-dimensional electrophoresis followed by identification mass spectrometry. PK measured a spectrophotometric assay. As controls, increases gene hexokinase, phosphoglucomutase, phosphofructokinase, aldolase triosephosphate isomerase found patients, but not T1DM(DN−) In protein analysis showed an altered three enzymes: triosophosphate isomerase, enolase PK. addition, lower than that conclusion, this study reports novel may be associated pathophysiology insulin resistance renal damage
