Development of an enzyme-linked immunosorbent assay for detection of circulating IgG autoantibodies against canine desmoglein 3 in dogs with pemphigus

摘要: Pemphigus, a group of autoimmune blistering dermatoses recognized in humans and dogs, is divided into three subgroups: pemphigus vulgaris (PV), foliaceus (PF) paraneoplastic (PNP). Previously, we have demonstrated that circulating anti-desmoglein 3 (Dsg3) IgG dogs with PV PNP disrupts Dsg3-mediated keratinocyte cell–cell adhesion. The aim this study was to develop an enzyme-linked immunosorbent assay (ELISA) for the detection against canine Dsg3 pemphigus. A secreted form recombinant protein representing entire extracellular domain (cDsg3) produced by baculovirus expression used as coated antigen ELISA. titre anti-cDsg3 ELISA significantly higher (n = 14) than healthy (n = 44) (P < 0.05). Of sera, 11/14 (79%) sera serum were positive On other hand, 14/37 (38%) from PF 9/18 (50%) subepidermal bullous Circulating cDsg3 not superficial pyoderma (n = 5), atopic dermatitis (n = 8), or (n = 44). Moreover, immunoreactivity all 21 completely absorbed preincubation cDsg3. These findings altogether suggest can be rapid screening test anti-Dsg3 autoantibodies pemphigus. Zusammengefasst weisen diese Ergebnisse darauf hin, rascher Screening Test fur Bestimmung zirkulierenden Anti-Dsg3 Autoantikorpern werden kann.
