摘要: A co-culture system based on encapsulated feeder cells within alginate beads was developed through optimizing the detailed aspects of cell culture to expand CD34-positive (CD34+) ex vivo. Mesenchymal stem isolated from different sources (human amniotic (hAMSCs) and umbilical cord (UCMSCs)), human fibroblast (HFs) have been respectively chosen as results showed that hAMSCs were superior UCMSCs HFs in conventional two-dimensional (2D) co-cultures regarding promotion total nucleated (TNCs) expansion maintenance CD34+ phenotype. Alginate employed limit growth hAMSCs, which could effectively restrict proliferation while viability still highly maintained. Intriguingly, only a few migrated out beads, whereas secreted factors cytokines be easily released. Furthermore, supported cells/hAMSCs 2D indirect exhibited increased TNCs expansion, higher percentages CD34+CD38− cells, better vitality when compared co-culture. Therefore, can promote
