Linoleic acid-induced endothelial cell injury: Role of membrane-bound enzyme activities and lipid oxidation

摘要: High plasma levels of linoleic acid (18:2) may injure endothelial cells, resulting in decreased barrier function the vascular endothelium. The effects on (transendothelial movement albumin), membrane-bound enzyme activities, and possible autooxidation under experimental conditions were studied. exposure monolayers to 18:2 for 24 hr at 60, 90, 120 microM fatty concentrations caused a significant increase transendothelial albumin, with maximum albumin transfer 90 microM. Fatty treatment resulted increased appearance cytosolic lipid droplets. Activities enzymes, angiotensin-converting (ACE), Ca(2+)-ATPase steadily increasing time cell 18:2, reaching significance hr. Treatment cultures up did not cause cytotoxicity, as evidenced by nonsignificant change cellular release [3H]-adenine. Incubation 18:2-supplemented serum-containing culture media 1000 37 degrees C 48 result formation products.
