摘要: Autologous haematopoietic progenitor cell (HPC) transplantation is increasingly used to restore haematopoiesis after high-dose chemotherapy treatments. The present study was designed analyse the ability of hydroxyapatite (HAP) seeded with endothelial cells (EC) support proliferation and differentiation CD34+ HPC in static culture conditions. HAP endothelializable as assessed by scanning electron microscopy time-course DNA synthesis analysis using tritiated thymidine incorporated EC isolated from human umbilical vein cord. Short-term coculture experiments which cord blood were on endothelialized HAP, performed. Results show that permissive expansion a maximum obtained between days 7 14 presence IL-1 IL-3 when compared other omitting either or interleukins. From morphological analyses, expanded population mainly belonged myelocytic lineage 33% mature (polymorphonuclear neutrophils monocytes) at day coculture. immature could remain trapped within while giving rise more progeny exit microenvironment.
