An in vivo and in vitro investigation into the effects of α-chlorohydrin on sperm motility and correlation with fertility in the han wistar rat

摘要: Abstract Following a recommendation from the International Conference on Harmonisation, pharmaceutical companies are now monitoring possible drug effects sperm motility in rat during preclinical safety studies by assessing (velocity). However, it is not known precisely how changes relate to fertility. Therefore, of α-chlorohydrin were investigated and related fertility both vivo vitro. α-Chlorohydrin was given orally male rats using range doses: 0, 5, 10, 20 mg/kg for at least 5 consecutive days. Sperm then assessed standard scoring system (operators' observation sperm) that graded degree (i.e., 0 = i mmotile 4 very motile). The results showed dose-related decrease motility. also appeared move with “jerky” action. Surprisingly, when this correlated fertility, none females mated treated males became pregnant. A pregnancy would perhaps have been expected. There no effect morphology, testicular epididymal pathology only seen after doses mg/kg.
