摘要: Increased thermal or mechanical stability of DNA duplexes is desired for many applications in nanotechnology -medicine where used as a programmable building block. Modifications pyrimidine bases are known to enhance and have the advantage standard base-pairing easy integration during chemical synthesis. Through single-molecule force spectroscopy experiments with atomic microscopy molecular assay we investigated effect pyrimidines harboring C-5 propynyl modifications on double-stranded DNA. Utilizing these complementary techniques, show that significantly increase if annealed at high temperature. In contrast, modified complexes formed room temperature short incubation times display same non-modified duplexes.
