摘要: The effect of 1 alpha,25-dihydroxyvitamin D3 (1,25-(OH)2D3) on pokeweed mitogen-induced generation human immunoglobulin-secreting cells (ISC) was studied in serum-free culture. Human peripheral blood mononuclear (1.75 X 10(6)/ml) were cultured 200 microliter microculture wells containing Iscove's MEM supplemented with 20 micrograms/ml transferrin, soybean lecithin and mg/ml delipidated bovine albumin. culture supported ISC all isotypes, the optimal concentration mitogen microgram/ml. After stimulation a suboptimal (0.3 micrograms/ml) mitogen, 1,25-(OH)2D3 suppressed at dose range between 10(-9) M 10(-7) M, maximal inhibition obtained M. had no spontaneous generation. specific for suppression by 24R,25-dihydroxyvitamin or 25-hydroxyvitamin D3.
