摘要: Total knee arthroplasty (TKA) is the most common and cost-effective treatment for older adults with long-standing osteoarthritis. During TKA, muscle cells suffer from prolonged oxygen deficiency, which leads to altered cell metabolism that reduces energy demand maintains homeostasis before blood flow restored. This study focused on role of lncRNA muscleblind-like 1 antisense RNA (MBNL1-AS1) in protecting sevoflurane-pretreated mice against ischemia-reperfusion (I/R) injury after as well elucidation potential associated mechanism. Identification differentially expressed lncRNAs was performed using microarray dataset GSE21164, extracted GEO database. Target genes were determined Multi-Experiment Matrix (MEM), a dual-luciferase reporter gene assay, KEGG enrichment analyses. The results showed MBNL1-AS1 overexpressed skeletal mice, while KCNMA1, enriched cGMP-PKG signaling pathway, negatively regulated by MBNL1-AS1. Furthermore, I/R displayed serious inflammatory reactions. Down-regulation increased expression PKGII, VASP, VEGF, Bcl-2, Cyclin D1, D3, Cdc 42 but decreased Bax, cleaved caspase-3, PARP. upon upregulation, rate apoptosis proliferation decreased. Our data suggested down-regulated might promote inhibit upregulating KCNMA1 via activation thus TKA.
