摘要: Although mRNA for the retinoic acid receptor alpha (RAR-alpha) is expressed in many different myeloid leukemias, most of these leukemia cells exhibit little if any phenotypic response when exposed to (RA). To determine whether such RA resistance related altered structure or function, we performed a detailed analysis nuclear receptors RA-resistant K-562 cells. These same approximate molecular weight and similar kd as those exhibited by RA-sensitive HL-60 cell line, but number (80 per cell) significantly lower than that RA- sensitive (550 cell). Retroviral-mediated transduction RAR-alpha cDNA into increased 2,000 cell. RAR-alpha-transduced cells, incubated with RA, diminished proliferation associated decreased c-myc expression an accumulation G0/G1. In addition, RA-treated downregulation CD15 surface antigen slight increase hemoglobin production manifest no other evidence significant erythroid, megakaryocytic, differentiation. results indicate elevated can be involved altering not necessarily differentiation certain treated
