摘要: Plant-based transient expression is an alternative platform to produce hemagglutinin-based subunit vaccines. This production system provides not only fast and effective response in the context of a pandemic but also enables supply big volume vaccines at low cost. Crude plant extracts containing influenza hemagglutinin are considered use as vaccine sources because avoidance related purification steps resulting cost allowing veterinary applications. Highly immunogenic hemagglutinins urgently required meet these pre-conditions. Here, we present new innovative way generate functional H5 oligomers from avian flu planta by specific interaction S·Tag S·Protein. A was fused trimers this construct transiently co-expressed with S·Protein-TPs which multimerized disulfide bonds via cysteine residues tailpiece sequences (TP) IgM antibody. Multimerized serve bridges/molecular docks combine S·Tag-fused form very large oligomers. crude extract were highly active hemagglutination high titers. Immunization mice two doses after storage for 1 week 4 °C caused strong immune responses induced neutralizing humoral mice. These results allow development cheap application future.
