摘要: The T cell activation is initiated by interaction of specific Ags with TCR, followed intracellular biochemical events leading to several genes. signal transducer and activator transcription (STAT) proteins in a primary TCR-mediated cells have been explored. In purified human peripheral blood cells, nuclear STAT were activated approximately 3 h after cross-linked anti-CD3 Abs. These detected using the IFN-gamma-activated sequence (GAS) related oligonucleotides as probes electrophoretic mobility shift assay. Analysis extracts anti-STAT Abs indicated that they contained STAT-3 additional crossreactive family. induction activity was inhibited completely pretreatment either cycloheximide or cyclosporin A, thus indicating due secondary factor produced cells. As neutralizing anti-IL-6 effectively down-regulated early exogenously added IL-6 rapidly DNA binding similar bindings, it can be concluded responsible for response.
