摘要: Background and Aims Colon cancer is the second leading cause of death due to worldwide. Elevated expression IGF-IR a frequent genetic abnormality seen in this malignancy. The aim study was examine anti-growth effects elicited by decrease protein level RNA interference (RNAi) SW480 cells. Methods A plasmid-based polymerase III promoter system used deliver express short interfering (siRNA) targeting reduce its IGF-1R detected Western blot. We assessed silencing on cell growth curve. Results prepared type hairpin (shRNA) vector that could efficiently inhibit At 48 h after transfection, inhibition rate 92 ± 2% at mRNA RT-PCR analysis. blotting similar levels cells transfected with pkD-shRNA-IGF-IR-V2. Downregulation resulted significant vitro . rates 24, 48, 72 pkD-shRNA-IGF-IR-V2 transfection were 32.06, 47.61, 35.36%, respectively. Conclusions Our data show decreasing RNAi significantly tumor , implying therapeutic potential treatment colon overexpression oncogenes such as IGF-IR. may be target for human cancer.
