摘要: Abstract The alkaline version of the single cell gel electrophoresis assay, popularly known as Comet is widely used to evaluate genotoxic potential chemicals and environmental contaminants, for monitoring purposes. In recent years, this assay has increasingly been recognized a potentially valuable tool regulatory studies. commonly utilises commercially available software programmes extent DNA damage at single-cell level. These provide large number measurement outcomes (i.e., tail length, %Tail DNA, various measures moment, etc.) migration damage. At however, there no general agreement with respect most relevant measurements or parameters use. This study was carried out establish which measurement(s) in are significantly correlated damage, should thus be adopted routine Pooled peripheral blood samples from 3 healthy human individuals were irradiated range doses 137Cs gamma-radiation (0, 1, 2, 4 8 Gy). Following irradiation, performed according standard protocol, different recorded by use Komet 5.0 (Kinetic Imaging Ltd., Liverpool, UK). correlation analysis, Olive Tail Moment (OTM), Extent percentage (%Tail DNA) gave good correlations that not each other. Further retrospective analysis other vitro vivo experiments chemical agents also suggested OTM dose used. Since many manuscripts, these two could continue applied However, since measured arbitrary units image-analysis systems give values, considered more meaningful easy conceptualise. Other might significant genotoxicological
