摘要: The fluid collected by direct leaf centrifugation has been used to study the proteome of sugar beet apoplastic as well changes induced Fe deficiency and resupply Fe-deficient plants in protein profile. Plants were grown Fe-sufficient conditions, was carried out with 45 μM Fe(III)-EDTA for 24 h. Protein extracts analyzed two-dimensional isoelectric focusing-SDS-PAGE electrophoresis. Gel image analysis revealed 203 consistent spots, proteins 81% them (164) identified nLC-MS/MS using a custom made reference repository sequences. When redundant UniProt entries deleted, non-redundant consisting 109 obtained. TargetP SecretomeP algorithms predicted that 63% secretory proteins. Functional classification indicated stress defense, metabolism, cell wall C metabolism accounted approximately 75% proteome. effects Fe-deficiency on apoplast limited, only five spots (2.5%) changing relative abundance, thus suggesting homeostasis is maintained upon shortage. identification three chitinase isoforms among increasing abundance suggests one few includes modifications. Iron deficient changed 16 when compared either or samples. Proteins these can be broadly classified those responding Fe-resupply, which included defense related proteins, non-responsive, are mainly whose followed same trend Fe-deficiency.
