Rheological and syringeability properties of highly concentrated human polyclonal immunoglobulin solutions

摘要: This study of highly concentrated polyvalent immunoglobulin solutions, IgG, aimed at analyzing the relationships between protein concentration and aggregation on one hand viscosity other hand. Viscosity variations as a function IgG showed two well-defined behaviours: Newtonian behaviour for low-concentrated solutions shear-thinning ones. The data fitted very well with Mooney model, suggesting absence intermolecular interactions in that behaved like non-interacting suspension hard particles. polyclonal nature seems to prevent interaction. shape factor, determined from fitting, revealed non-spherical molecules. rheological properties were also correlated injection force (F) through hypodermic needles by syringeability tests. Here, F was mainly affected three parameters: solution viscosity, flow rate, needle characteristics. In fact, tests increased rate decreased internal diameter needle. A zone optimal conditions then identified taking into account different affecting parameters maximum manual injection, which fixed 30N.
