Relationship between somaclonal variation and type of culture in cucumber

摘要: Highly inbred B line of cucumber was used to compare the effect four types in vitro culture on somaclonal variation. The plants were regenerated from following culture: twelve- and eighteen-month-old liquid meristematic clumps (LMC12(18)), ten-month-old embryogenic cytokinin-dependent suspension (CDS), medium with modified NH+ 4/NO3 - ratio (CDS 1.7), twelve-month-old auxin-dependent (ADS), thirty six-month-old (ADS 1.7) recurrent leaf callus regeneration (RLC) – repeated 5 times. differences incidence properties observed: ploidy R0 plants, segregation new morphological traits R1 germination ability seeds. families phenotypes most numerous CDS (62.5%) LMC18 (57.9%), next CDS1.7 (35.7%), while smallest number found LMC12 (11.1%) RLC (3.4%).Tetraploid mixoploid occurred ADS1.7 ADS (100%) whereas observed contain only tetraploids, respectively 33.3% 55.2%. There no changes after LMC12, CDS1.7. Among there such that have not been described so far cucumber: ginkgolike (gll), yellow-green chlorophyll mutants (y-gc), serrate margin corolla male female flowers (smc).
