An analysis of the proteomic profile for Thermoanaerobacter tengcongensis under optimal culture conditions.

摘要: The genome of Thermoanaerobacter tengcongensis is estimated to encode 2588 theoretical proteins. In this study, we have vitalized approximately 46% the proteome experimentally using a proteomic strategy that combines three different methods, shotgun digestion plus high-performance liquid chromatography (HPLC) with ion-trap tandem mass spectrometry (shotgun-liquid (LC)/MS), one-dimensional sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) HPLC (one-dimensional (1DE)-LC/MS), and two-dimensional matrix-assisted laser desorption/ionization-time flight-mass (2DE-MALDI-TOF-MS). Of 1200 proteins identified, as few 76 were globally found by all approaches, notably, most these in soluble fraction. However, there number unique detected one method only, suggesting our provides means toward obtaining comprehensive view protein expression profile. Proteins from major metabolic pathways are strongly represented on map, enzymes identified more than method. Based upon present able broaden understanding how T. survives under high temperature environment, whereas several its properties can not be fully explained data.
