Constitution of semen samples from XYY and XXY males as analysed by in-situ hybridization*

摘要: A brightfield microscopical in-situ hybridization (ISH) technique was applied to semen samples of two 47,XYY males, one 46,XY/47,XXY/XXY male with fertility problems, and normal 46,XY men, who served as controls. The use a standardized nuclear DNA decondensation method, together double-target ISH morphological staining, allowed an accurate study the sex chromosomal content morphology spermatozoa. In males carrying extra chromosome, we detected X- Y-bearing spermatozoa in ratio which did not differ significantly from 1:1 found males. Aneuploidy for chromosomes approximately 15% both XYY 3% XXY male. most striking finding relatively low percentage these patients, average 65% 84% other cells represented immature germ (IGC), including spermatogonia spermatocytes arrested at various stages spermatogenesis. Apparently, or IGC are shed into increased extent compared normal, fertile men. chromosome constitution heterogeneous. However, that majority 47,XXY carried single strengthens hypothesis cell line must be present, apparently proliferative advantage over cells.
