DNA fingerprinting of bluegill sunfish (Lepomis macrochirus) using (GT)n microsatellites and its potential for assessment of mating success

摘要: Variable microsatellite DNA markers are powerful tools to study parentage and gene flow in fish populations. In this work, we cloned characterized 60 (GT) repeat sequences from the bluegill sunfish (Lepomis macrochirus) genome. We report frequency distribution, lengths, proportions of classes for comparison with microsatellites isolated other teleost, mammal, invertebrate genomes. Bluegill similar those fish, strengthening previous reports significant differences organization between teleosts mammals. Primers polymerase chain reaction amplification were designed 17 loci. The allele frequencies, degree polymorphism, heterozygosities estimated using Lake Opinicon, Ontario. Seven loci potentially highly informative fingerprinting studies parentage. A preliminary paternity analysis two these on brood a natural nest revealed their potential utility at estimating relative success reproductive life histories, parental cuckolder, characteristic male bluegill. Cross-amplification related species showed some locus conservation among taxa, allowing variety centrarchids.
