摘要: We have developed a new, quick and efficient method of high-performance liquid chromatography (HPLC) for the isolation quantitative determination phospholipids in hepatocyte membranes. A silica gel column was used determination, an isocratic mixture acetonitrile, methanol 85% phosphoric acid (130:5:1.7, v/v/v) as mobile phase. Six kinds phospholipids, i.e. phosphatidylinositol (PI), phosphatidylserine (PS), phosphatidylethanolamine (PE), phosphatidylcholine (PC), lysophosphatidylcholine (LPC) sphinogomyelin (SPH), this order, were completely isolated within 45 min. The phospholipid composition sinusoidal membrane vesicles (SMV) canalicular (CMV) obtained from rat liver, well human erythrocyte ghosts determined by HPLC method. level SPH CMV significantly higher than that SMV, PC lower SMV. These results considered attributable to low fluidity CMV. different SMV present technique is suitable cell
