Experimental design optimization of the capillary electrophoresis separation of leucine enkephalin and its immune complex.

摘要: To optimize the capillary electrophoretic separation conditions for leucine enkephalin (LE) and immune complex of LE anti-LE reaction, an analysis using a three-level, three-factorial Box-Behnken design was performed. Three parameters, buffer pH (X(1)), concentration (X(2)), applied voltage (X(3)), were chosen to observe effect on responses. The responses theoretical plate number, migration time peak, resolution between peaks. optimum process validation determined statistical regression surface plot diagrams. electrophoresis established be 75 mM phosphate at 7.00 with 15 kV. By technique, prediction satisfactory verification yielded values within +/-5% range predicted efficiency.
