摘要: Lazaroids are potent inhibitors of lipid peroxidation. Endothelial cell damage has been shown to occur during cold storage preservation lung and liver. This study examines the effects on endothelial viability addition four lazaroids, U74006F, U78518F, U74500A, or U75412E solutions. Human umbilical vein cultures were stored at 4 degrees C for 48 96 hr in EuroCollins 5% polyethylene glycol buffered saline (PEG). U75412E, dexamethasone (each 50 microM) was added EC (n = 32) PEG compared with control solutions alone. determined by measuring cellular reduction 3-[4,5-dimethylthiazol-2-yl]-2,3-diphenyltetrazolium bromide a purple formazan dye. The occurs only viable cells requires mitochondrial dehydrogenase activity. Results quantified dye absorbance (Ab) micro-ELISA spectrophotometer. Absorbance values ANOVA reported as mean +/- standard deviation. Addition U74500A (Ab 0.474 0.055) 0.462 .005) improved when 0.289 0.069) 0.287 0.052) alone (P less than 0.05). At hr, resulted both 0.377 0.068) 0.195 0.09) 0.212 0.1) Other lazaroids tested also effective improving viability, but lesser degree U74500A. demonstrates that organ improves viability.
